Heterochromatin is one of the two forms of chromatin, the other being euchromatin. Chromatin is the complex of DNA and proteins that makes up chromosomes within the nucleus of eukaryotic cells. Heterochromatin is characterised by its dense packing, which makes it appear darker under a microscope compared to the less condensed euchromatin. This compact structure is achieved through a higher level of histone modifications and DNA methylation, which are chemical changes that help to wind the DNA more tightly around the histone proteins.
The primary function of heterochromatin is to regulate gene expression. Because of its compact structure, the genes within heterochromatin are generally inaccessible to the machinery that transcribes DNA into RNA, which is the first step in gene expression. This means that these genes are typically silent or expressed at low levels. However, this silencing is not permanent and can be reversed under certain conditions, allowing the genes to be expressed when needed.
Heterochromatin also plays a crucial role in maintaining chromosome stability. The ends of chromosomes, known as telomeres, and the points where sister chromatids are joined, known as centromeres, are both composed of heterochromatin. The compact structure of heterochromatin at these locations helps to protect the DNA from damage and ensures the correct segregation of chromosomes during cell division.
In addition, heterochromatin is involved in DNA replication. During the S phase of the cell cycle, when DNA is replicated, heterochromatin is replicated later than euchromatin. This late replication is thought to help maintain the integrity of the DNA sequence within heterochromatin, as it allows more time for any errors that occur during replication to be corrected.
