The Kozak sequence is a specific sequence of nucleotides in the mRNA that is recognised by the ribosome during the initiation of translation, the process by which the genetic code in mRNA is read to synthesise a protein. This sequence is named after Marilyn Kozak, the scientist who discovered its role in translation initiation.
The Kozak sequence is located upstream of the start codon (AUG) on the mRNA. It is not the same in all organisms, but in mammals, it is typically (gcc)gccRccAUGG. The R represents a purine base (adenine or guanine), and the underlined AUG is the start codon where translation begins. The bases in parentheses are less conserved, meaning they can vary more between different mRNA molecules.
The ribosome, a large complex of RNA and proteins, is responsible for translating the mRNA into a protein. It must first bind to the mRNA at the correct location to start translation at the right place. The Kozak sequence helps to guide the ribosome to the correct start site. The ribosome recognises the Kozak sequence and positions itself so that the AUG start codon is in the P site, the location in the ribosome where the first tRNA molecule binds to start protein synthesis.
The Kozak sequence is crucial for efficient translation initiation. If the sequence is mutated or not optimal, the ribosome may not bind correctly, leading to inefficient translation or even translation starting at the wrong codon. This could result in a protein with a different sequence of amino acids, potentially affecting its function.
